Saddest: Joyce's death, Buffy's Death, Tara's "illness",
Funniest: here the Funniest Actions Award goes straight to Xander - he's split moments, he's servantry to Dracula, He's treatment to Dawn, etc, etc, etc.
Important: Buffy is ready to sacrifuce herself to save the world - her merely first major act or maturity. Willow's able to fight for her love, and show that she's strong.
I'd think of more later..

Hmm.....
Saddest: Joyce's death
Funniest: Xander as the 'spider eating man bitch' aka Dracula's Butt Monkey. :-D. He even had the crazy servant walk down
Most Impowering: Buffy's death. The jump, the speech, the aftermath, the whole shebang

Saddest - Joyce's death. I think that's pretty much undisputed at the moment!
Funniest - Xander doing the 'Snoopy Dance' all wet, just to prove to Willow that he is who he claims to be, forgetting that the other Xander could have done the same!
Most Impowering - When Buffy stands up to Tara's family, claiming they'll have to go through her. Sweet, impowering and just totally Buffy. Plus it was like complete acceptance of Tara, despite the mixed feelings towards her at the start of that episode!

Well, I'll dispute it... "The Body" didn't touch me as I didn't mind Joyce dying (I know... that opinion probably doesn't mark me as a nice person, but that's how I felt). I have to admit though, having just recently rewatched the entire S5, that this ep definitely has one big point in its favor: natural causes, a real-life situation without anything supernatural in it (if you leave out the vamp in the morgue, that was hardly more than "ok, we know this show's about vampires, so here's one for you guys who absolutely can't do without'em"). This was a major issue in why I disliked both "Ted" and "Gingerbread", both of which could have been milestones of the show if they had left out the demon and the robot.
So, here's my list:
Saddest - Tara getting brainsucked by Glory (even more intense as it happened immediately after her argument with Willow)
Most empowering - the Scoobies standing up for Tara at the end of Family
Funniest - Xander as Dracula's "Renfield", and pretty much every single second with the Buffybot
